GenisteinGenistin is an isoflavone found in a number of dietary plants like soy and kudzu. It was first isolated in 1931 from the 90% methanol extract of a soybean meal, when it was found that hydrolysis with hydrochloric acid produced 1 mole each of genistein and glucose. Chemically it is the 7-O-beta-D-glucoside form of genistein and is the predominant form of the isoflavone naturally occurring in plants. In fact, studies in the 1970s revealed that 99% of the isoflavonoid compounds in soy are present as their glucosides. The glucosides are converted by digestive enzymes in the digestive system to exert their biological effects. Genistin is also converted to a more familiar genistein, thus, the biological activities including antiatherosclerotic, estrogenic and anticancer effects are analogous. Genistein, an isoflavone, is a compound derived mainly from soy. Various anticancer studies have demonstrated genistein to affect the process of tumorigenesis/carcinogenesis mediated via epigenetic regulations (Zhang and Chen, 2011). Reports are suggestive of possible role of genistein in inhibition of DNA methyltransferases (DNMTs) that could regulate gene expression by preventing DNA methylation at promoter levels. Genistein has also proved to reduce the risk of breast cancer and enhances DNA demethylation of SF1 promoter in endometrial stromal cells present in the endometrium (Khan et al., 2011). Genistein also reduces hypermethylation levels of tumor suppressor genes such as CHD5 and promotes the expression of CHD5 as well as p53 that prevents neuroblastoma (NB) growth and formation of tumor microvessel in vivo (Li et al., 2012). In addition to these, genistein could also act as an inhibitor to reduce the expression of DNMT3b in NB model, thereby paving its way as an adjuvant therapeutic candidate for NB treatment.
Supplement Dosages
Although isoflavones are mostly consumed by eating foods that contain them, there are isoflavone supplements in capsule form that are taken by mouth. The following doses of isoflavones are backed by clinical research studies: For postmenopausal individuals: A supplement with at least 54 mg of genistein (a soy isoflavone) per day is suggested for hot flashes. For IBS: Take a supplement of 40 mg of isoflavones per day for six weeks. For protection against osteoporosis: A supplement of 80 mg per day of soy isoflavones was associated with a dosage that reduced bone loss in postmenopausal individuals (protecting against osteoporosis). For general supplementation: There is no recommended dose established for general use.
